These electrode holders are designed exclusively for use the with MICRO-ePORE™ system, which includes the silver wire electrode holders. The platinum wire electrode holders are an optional accessory. Platinum wire electrode holders are important when you are working with proteins. The silver chloride wire is perfect for work with RNA. However, the silver chloride wire tends to denature proteins, reducing the efficiency of the microinjection solution.

*Note: Please do not attempt to take a Femtojet-purchased handle and screw the ePORE plastic head in, the threads are different and will damage the component.

Applications for Silver Wire Electrode Holders

Silver wire electrodes are suitable for most applications.

Applications for Platinum Wire Electrode Holders

Initially CRISPR-Cas9 technology started with RNA antisense sequences, but it is now moving towards proteins. Using platinum electrodes offers more active insertion of the genetic material capability, and there is a better chance of changing the genome. Some common protein applications for the platinum wire electrode holders include:

  • crRNA contains the guide RNA that locates the correct segment of host DNA along with a region that binds to tracrRNA (generally in a hairpin loop form), forming an active complex.
  • tracrRNA binds to crRNA and forms an active complex.
  • sgRNA (single-guide RNA) is a combined RNA consisting of a tracrRNA and at least one crRNA.
  • Cas9 (most common) involves an enzyme whose active form is able to modify DNA. Many variants exist with different functions (i.e. single-strand nicking, double-strand breaking, DNA binding) due to each enzyme's DNA site recognition function.
  • Repair Template is a DNA molecule used as a template in the host cell's DNA repair process, allowing insertion of a specific DNA sequence into the host segment broken by Cas9.
